【摘要】  目的:觀察麥冬治療妊娠糖尿病胰島素抵抗的作用,研究其作用機(jī)制. 方法:選妊娠10 d大鼠,建立妊娠糖尿病胰島素抵抗動(dòng)物模型,正常大鼠為對(duì)照組,隨機(jī)將40只妊娠糖尿病胰島素抵抗大鼠分為4組(n=10):GDM組、GDM+麥冬組、GDM+羅格列酮和GDM+羅格列酮+麥冬組. 觀察用藥前后血糖、胰島素的變化,檢測(cè)腹腔脂肪組織中TNFα與leptin的mRNA表達(dá)變化. 結(jié)果:與對(duì)照組比較,GDM組大鼠空腹及餐后血糖與胰島素較正常對(duì)照組明顯升高(P<0.05),TNFα與leptin的mRNA表達(dá)明顯增加(P<0.05);與GDM組比較,麥冬治療組、羅格列酮治療組、羅格列酮+麥冬治療組大鼠空腹及餐后血糖均下降(P<0.05),TNFα與leptin的mRNA表達(dá)明顯降低(P<0.05);與麥冬治療組比較,羅格列酮治療組空腹及餐后血糖無明顯變化(P>0.05),TNFα與leptin的mRNA表達(dá)無明顯變化(P>0.05),羅格列酮+麥冬治療組空腹及餐后血糖明顯下降(P<0.01),TNFα與leptin的mRNA表達(dá)明顯降低(P<0.05);與羅格列酮組比較,羅格列酮+麥冬治療組空腹及餐后血糖明顯下降(P<0.01),TNFα與leptin的mRNA表達(dá)明顯變化(P<0.05). 結(jié)論:麥冬治療妊娠期糖尿病療效明顯,能變胰島素抵抗?fàn)顟B(tài),其機(jī)制與TNFα與leptin的mRNA表達(dá)的變化有關(guān)醫(yī).學(xué)全.在.線網(wǎng)站m.zxtf.net.cn.

Treatment effect of Chinese herb Maidong on gestational diabetes mellitus complicated by insulin resistance and its underlying mechanism

【Abstract】 AIM: To investigate the treatment effect of Chinese herb Maidong on gestational diabetes  mellitus (GDM) and insulin resistance (IR) and to study its underlying mechanism. METHODS: GDM complicated by IR was induced in rats of 10 gestation days. Normal rats served as a control group. Forty model rats were randomized into GDM group, GDM & Maidong group, GDM & rosiglitazone maleate group, GDM, Maidong & rosiglitazone maleate group (n=10). Serum glucose and serum insulin were determined after and before administration. The expressions of TNFα and leptin were detected in abdominal adipose tissues after and before administration. RESULTS: Compared to controls, serum glucose and serum insulin were evidently increased (P<0.05) and the expressions of TNFα and leptin also significantly rose (P<0.05) in GDM group. Compared to GDM group, serum glucose and serum insulin were significantly decreased (P<0.05) and the expressions of TNFα and leptin declined obviously, too (P<0.05) in GDM & Maidong group, GDM & rosiglitazone maleate group, GDM & Maidong & rosiglitazone maleate group. Compared to GDM & Maidong group, serum glucose and serum insulin remained unchanged (P>0.05)and the expressions of TNFα and leptin were also unchanged (P>0.05) in rosiglitazone maleate group, while serum glucose, and serum insulin (P<0.01) and the expressions of TNFα and leptin were signicantly reduced (P<0.05) in Maidong & rosiglitazone maleate group. Compared to rosiglitazone maleate group, serum glucose and serum insulin were significantly decreased (P<0.05) and the expressions of TNFα and leptin were unchanged (P<0.05) in Maidong & rosiglitazone maleate group. CONCLUSION: Treatment effect of Chinese herb Maidong is good for GDM and can reverse IR. These actions are related to the changes of the contents of TNFα and leptin.
